چکیده

The relative rates of tRNA, rRNA, and DNA synthesis were measured for steady state cultures of Escherichia coli B/r growing in succinate and glucose minimal media which support growth rates of 0.67 and 1.46 doublings per hour, respectively. Following a Z-fold increase in growth rate, the rate of tRNA synthesis increased approximately Z-fold relative to the rate of DNA synthesis and decreased 10 to 15 % relative to the rate of rRNA synthesis. It is concluded that the synthesis of tRNA is regulated in a manner similar to the mechanism regulating rRNA synthesis. The 10 to 15% difference in the relative rates of synthesis is not the result of an increase in the relative frequency of ribosomal RNA cistrons at the higher growth rates, and therefore it seems that the mechanisms controlling rRNA and tRNA synthesis are at least partially independent.

Stability of ribosomal and transfer ribonucleic acid in Escherichia coli B/r after treatment with ethylenedinitrilotetraacetic acid and rifampicin.

A short treatment with ethylenedinitrilotetraacetic acid to permeabilize bacteria for various antibiotics or treatment with the ribonucleic acid (RNA) synthesis inhibitor rifampin causes a slow degradation of 50S and 30S ribosomal particles and of the corresponding 23S and 16S ribosomal RNA species (about 25 percent in 1 h). The effects are additive such that the decay is about 50 percent/h if ...

Synthesis and function of ribonucleic acid polymerase and ribosomes in Escherichia coli B/r after a nutritional shift-up.

The syntheses of stable ribosomal ribonucleic acid (RNA) and transfer RNA in bacteria depend on the concentration and activity of RNA polymerase and on the fraction of active RNA polymerase synthesizing stable RNA. These parameters were measured in Escherichia coli B/r after a nutritional shift-up from succinate-minimal to glucose-amino acids medium and were found to change in complex patterns ...

Isoleucine and threonine can prolong protein and ribonucleic acid synthesis in pyridoxine-starved mutants of Escherichia coli B.

Pyridoxineless mutants of Escherichia coli B stopped incorporation of nucleosides into trichloroacetic acid-insoluble material about 40 to 60 min after pyridoxine starvation was initiated, whereas incorporation of amino acids (measured the same way) slowed but did not stop for several hours. Both these incorporations and cell density were increased most effectively by the presence of either thr...

Deoxyribonucleic acid-ribonucleic acid hybridization studies on the L-Arabinose operon of Escherichia coli B-r.

An increase in the rate of synthesis of ara-specific messenger ribonucleic acid as measured by deoxyribonucleic acid-ribonucleic acid hybridization has been detected in the induced wild-type (ara(+)) strain of Escherichia coli B/r as compared with the uninduced control, thus providing evidence that regulation of the positively controlled l-arabinose operon is at the level of transcription.

Regulation of ribosomal protein synthesis in Escherichia coli.

The kinetics of synthesis of ribosomal, nonribosomal, and total protein, and of ribosomal ribonucleic acid (RNA), were measured in Escherichia coli during a shift-up involving a doubling of specific growth rate. The increase in ribosomal protein synthesis was not accompanied by a corresponding decrease in non-ribosomal protein synthesis. Thus, the average rate of protein synthesis per ribosome ...
